XTT Cell Proliferation and Cytotoxicity Assay Kit uses 2,3-bis(2-methoxy-4-nitro-5-sulfophenyl)-5-
[(phenylamino)carbonyl]-2H-tetrazolium hydroxide is a colorimetric assay for rapid, high-sensitivity detection of cell proliferation and cytotoxicity. In the presence of an electron-coupling agent, XTT is reduced by NAD(P)H-dependent dehydrogenases in mitochondria to form an orange-yellow, water-soluble Formazan. The amount of Formazan generated is proportional to the number of viable cells. Greater or faster cell proliferation results in a darker color, while higher cytotoxicity yields a lighter color. For the same cell type, color intensity correlates linearly with cell count.
