Hematoxylin is a natural dye extracted from logwood and widely used in various formulations—such as Mayer's, Gill's, and Harris's—to stain cell nuclei, mitochondria, mucins, elastic fibers, muscle, collagen, myelin sheaths, and phospholipids. Its staining mechanism relies on the binding of positively charged basic hematoxylin to negatively charged acidic substances, resulting in a blue coloration.
Eosin (also known as erythrosine) is a red fluorescent dye that dissociates in water into negatively charged anions. These anions bind to positively charged cations in protein amino groups, staining cell cytoplasm, red blood cells, collagen, muscle fibers, connective tissue, and eosinophilic granules various shades of red or pink. As an acidic dye, it is commonly used in Hematoxylin and Eosin (H&E) staining as a counterstain for hematoxylin.
