Cell Counting Kit-8 (CCK-8) enables rapid detection using the water-soluble tetrazolium salt WST-8 (chemically named: 2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazole monosodium salt). In the presence of an electron-coupling reagent, WST-8 is reduced by mitochondrial dehydrogenases to produce a yellow-orange formazan dye.

Cell Counting Kit-8 (CCK-8) enables rapid detection using the water-soluble tetrazolium salt WST-8 (chemically named: 2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium monosodium salt). In the presence of an electron-coupling reagent, WST-8 is reduced by mitochondrial dehydrogenases in viable cells to produce an orange-yellow formazan dye. Colorimetric measurement allows for dynamic quantification of living cell numbers, enabling assessment of cell proliferation or drug toxicity. CCK-8 solution can be added directly to cells without pre-mixing with other components, offering simple operation and accurate results.

Cell Counting Kit-8 (CCK-8) is a redox indicator that, in the presence of the electron carrier 1-Methoxy PMS, uses dehydrogenases in live cells to catalyze the conversion of tetrazolium salt WST-8 into a formazan dye. The amount of formazan produced is linearly proportional to the number of viable cells.
